Autoreactivity of Peripheral Helper T Cells in the Joints of Rheumatoid Arthritis

Abstract Autoreactive CD4 T cells are thought to play pivotal roles in the pathogenesis of rheumatoid arthritis (RA). Recently, a subset that express high levels programmed death-1 (PD-1) but distinct from follicular helper have been identified joints RA patients and named peripheral (Tph) cells. Because PD-1 is expressed on chronically stimulated with Ags, we tested hypothesis Tph pathogenic autoreactive RA. We found human produce proinflammatory effector cytokines, including IFN-γ, TNF-α, GM-CSF, addition B cell–helping such as IL-21 CXCL13. Flow cytometric analysis showed different bias TCR Vβ usage between PD-1high PD-1low/neg cells, Th1 joint or memory blood, whereas there was little difference latter two subsets. In line this, deep sequencing demonstrated an overlap expanded clones blood not joint.
